تأثیر ضدقارچی عصاره‌ی متانولی حنا بر سویه‌های کاندیدا آلبیکانس جدا شده از بیماران مبتلا به کاندیدیازیس دهان

چکیده

مقدمه: داروهای ضد قارچ متعددی برای درمان کاندیدیازیس دهانی ارائه شده که هر کدام مزایا و عوارضی دارد. استفاده از گیاهان دارویی به عنوان عامل ضد قارچی، می‌تواند به دلیل کاهش عوارض ناشی از مصرف دارو کمک کننده باشد. هدف از مطالعه، بررسی تأثیر ضد قارچی عصاره‌ی متانولی حنا بر سویه‌های کاندیدا آلبیکانس بود.
مواد و روش‌ها: در این مطالعه‌ی تجربی- آزمایشگاهی، یک سویه استاندارد و ۵ سویه بالینی قارچ کاندیدا آلبیکانس، از ۵ بیمار مبتلا به بیماری کاندیدیازیس دهانی جدا شد. این مطالعه در سال 1402 در دانشگاه آزاد اسلامی واحد اصفهان (خوراسگان) انجام شد. سلول‌های کاندیدا کشت داده شد و سوسپانسیون‌های قارچی استاندارد و معادل با غلظت نیم مک‌فارلنز آماده و بر سطح محیط کشت سابارو دکستروز آگار کشت گردید. مقدار ١٠٠ میکرولیتر از عصاره آماده شده با غلظت‌های mg/ml ٦/٠، ٢٥/١، ٥/٢، ٥، ١٠ به چاهک اضافه و انکوبه شدند. میزان ١٠٠ میکرولیتر از رقت‌هایی از عصاره‌ی حنا به چاهک‌های میکروپلیت اضافه شد و قطر هاله عدم رشد و کدورت ناشی از رشد اندازه‌گیری و مقایسه گردید. داده‌ها با آزمون‌های آماریMann Whitney  و Kruskal-Wallis و ANOVA آنالیز شدند (0/05 = α).
یافته ها: در مقدار کدورت ناشی از رشد بین غلظت‌های مختلف عصاره‌ی حنا، نیستاتین و کنترل مثبت در سویه‌های کاندیدا آلبیکنس، تفاوت معنی‌دار وجود داشت (0/05 > p value) در مقدار درصد مهار رشد بین غلظت‌های مختلف عصاره، نیستاتین و کنترل مثبت در سویه‌های کاندیدا آلبیکنس، تفاوت معنی‌دار وجود داشت (0/05 > p value).
نتیجه گیری: تأثیر ضدقارچی عصاره‌ی حنا در غلظت‌های 1/25، 2/5، 5 و 10 بر علیه کاندیدا آلبیکنس جدا شده از بیماران مثبت بود.

Antifungal Effect of Henna Methanolic Extract on Candida Albicans Strains Isolated from Patients with Oral Candidiasis

Introduction: Several antifungal drugs have been proposed for the treatment of oral candidiasis, each of which has advantages and side effects. The use of medicinal plants as an antifungal agent can be helpful due to the reduction of side effects caused by drugs. This study aimed to determine the antifungal effect of methanolic extract of henna on Candida albicans strains.
Materials and Methods: In this experimental laboratory study, one standard strain and five clinical strains of Candida albicans were isolated from five patients with oral candidiasis. This study was conducted in 2023 at Isfahan Islamic Azad University. Cultured candida cells were prepared, and standard fungal suspensions with a concentration of 0.5 McFarland were prepared and cultured on the surface of the sabouraud dextrose agar culture medium. To the wells, 100 microliters of the prepared extract with concentrations of 0.6, 1.25, 2.5, 5, and 10 mg/ml were added, and the plates were incubated. To the wells of the microplate, 100 microliters of henna extract dilutions were added. Then, the diameter of the microorganism Non-Growth halo and turbidity caused by growth was measured and compared. Data were analyzed with Mann-Whitney, Kruskal-Wallis, and ANOVA statistical tests (α = 0.05).
Results: A Significant difference in the amount of turbidity caused by growth between different concentrations of extract, nystatin, and positive control in strains of Candida albicans was observed (p value < 0.05). There was a significant difference in growth inhibition percentage between different concentrations of extract, nystatin, and positive control in strains of Candida albicans (p value < 0.05).
Conclusion: The antifungal effect of henna extract in concentrations of 1.25, 2.5, 5, and 10 mg/ml was positive against Candida albicans isolated from patients.
